二型固有淋巴细胞(ILC2s)是近期被报道有助于2型炎症、组织修复和调节代谢平衡的先天免疫细胞。ILC2s 在各种生物作用中扮演着重要的角色,由于这类细胞比较稀少,使得对其表型和功能分析存在一定技术难度。这次网络研讨会将会讨论用于检测小鼠模型和人体组织动态细胞群的前沿技术,集中评估了前列腺素D2受体,CRTH2Th2细胞表达的化学引诱物同源分子)的作用,前列腺素D2受体和CRTH2ILC2免疫应答里用强化选择的技术方法来分析ILC2的表型和功能。这种技术包括多色流式细胞术,细胞分选处理,单细胞RNA染色(PrimeFlow™ RNA assay)和ILC2体外功能分析。另外,我们会介绍新推出的产品PrimeFlow™ RNA assay,它能在单细胞水平同时检测细胞中的RNA和蛋白质。这次会议我们将会为研究ILC2的研究人员提供ILC领域的相关信息。

Group 2 innatelymphoid cells (ILC2s) are recently described innate immune cells thatcontribute to type 2 inflammation, tissue repair, and metabolic homeostasis inmultiple tissues. While ILC2s play key roles in various biological processes,these cells are rare, making analysis of their phenotype and function atechnical challenge. This webinar will discuss cutting-edge techniques that canbe used to examine this dynamic cell population in murine models and in humantissues. A project focused on assessing the role of the prostaglandin D2receptor, CRTH2 (chemoattractant homologous molecule expressed by Th2 cells),in ILC2 responses will be used to highlight selected technical approaches andassays that can be used to analyze ILC2 phenotype and function. Techniques thatwill be highlighted include multi-color flow cytometry, cell sortingapproaches, single-cell RNA staining, and in vitro assays for ILC2 function. Inparticular, the PrimeFlow™ RNA assay, which enables simultaneous assessment ofprotein and RNA transcript expression on a single cell level, will bediscussed. The key concepts described during the session will provide relevantinformation for those new to the ILC field and for investigators who arealready experienced in the analysis of ILC responses.
